基于网络药理学探讨党参治疗2型糖尿病的作用机制

摘要:
目的:基于网络药理学方法探讨党参治疗2型糖尿病(T2DM)的潜在作用机制。方法:通过中药系统药理学分析平台数据库(TCMSP)及OMIM、TTD、Drugbank等网络数据库分别获取药物有效成分、活性靶点、疾病靶点,并取药物成分和疾病的交集靶点,构建药物活性成分与关键靶点网络、蛋白质相互作用(PPI)网络,并对靶蛋白进行GO富集分析和KEGG通路分析。结果:得到药效活性成分21种,药物靶点108个,T2DM靶点2402个,两者匹配映射得到关键靶点28个。党参治疗T2DM的主要成分为木樨草素、7-甲氧基-2-甲基-异黄酮、豆甾醇、黄豆黄素等,关键靶点为白细胞介素-6(IL-6)、血管内皮生长因子A(VEGFA)、过氧化物酶体增殖物激活受体r(PPARG)等,关键的生物学进程包括药物反应、葡萄糖稳态;关键通路为PI3K-Akt 信号通路、AMPK信号通路、FoxO信号通路、胰岛素抵抗。结论:党参治疗T2DM具有多成分、多靶点、多途径的特点。

Abstract:
Objective:To investigate the mechanism of action of Codonopsis pilosula in the treatment of type 2 diabetes mellitus (T2DM) based on network pharmacology.Databases including Traditional Chinese Medicine Systems Pharmacology,OMIM,TTD,and Drugbank were used to obtain the effective constituents and active targets of the drug and related disease targets,and the intersecting targets of drug constituents and the disease were obtained to construct an active component-key target network and a protein-protein interaction (PPI) network.Gene ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis were performed for target proteins.Results:A total of 21 active components,108 drug targets,and 2402 T2DM targets were obtained,and 28 key targets were obtained after mapping.The main components of Codonopsis pilosula in the treatment of T2DM included luteolin,7-methoxy-2-methyl isoflavone,stigmasterol,and glycitein;the key targets included interleukin-6,vascular endothelial growth factor A,and peroxisome proliferator-activated receptor γ;the key biological processes included drug reaction and glucose homeostasis;the key pathways included the PI3K-Akt signaling pathway,the AMPK signaling pathway,the FoxO signaling pathway,and the insulin resistance signaling pathway.Conclusion:Codonopsis pilosula has the features of multiple components,targets,and pathways in the treatment of T2DM.
