基于脂质组学的蛭龙活血通瘀胶囊抗动脉粥样硬化作用机制研究

摘要:
目的:基于脂质组学观察蛭龙活血通瘀胶囊抗动脉粥样硬化的作用机制。方法:将34只大鼠分为空白组、模型组、给药组,通过大鼠一次性腹腔注射维生素D3联合高脂饲料建立大鼠动脉粥样硬化模型。测定各组大鼠血脂[总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白、高密度脂蛋白(HDL-C)]水平,观察各组主动脉的病理变化,采用UPLC-Orbitrap MS 技术对各组血浆样本进行脂质组学研究。结果:与空白组相比,模型组TC、TG、LDL-C水平升高,差异具有统计学意义(P<0.05或P<0.01);与模型组相比,给药组 TC、TG、LDL-C水平降低,差异具有统计学意义(P<0.01)。组织病理学显示,给药组脂质沉积减少,主动脉内膜增厚改善。鉴定出脂质亚类29种,其脂质分子 959种。给药组与模型组差异脂质分子62种,主要差异代谢物为甘油磷脂、鞘脂及甘油酯类。在3组差异代谢物中,筛选出符合给药组趋势与模型组相反的差异性代谢物22种。结论:蛭龙活血通瘀胶囊可降低血脂及改善动脉粥样硬化,其主要作用通路为甘油磷脂代谢及甘油酯代谢。

Anti-atherosclerotic mechanism of Zhilong Huoxue Tongyu capsules based on lipidomics

Abstract:
Objective:To investigate the anti-atherosclerotic mechanism of Zhilong Huoxue Tongyu capsules based on lipidomics.Methods:A total of 34 rats were divided into blank group,model group,and administration group,and a rat model of atherosclerosis was established by a single intraperitoneal injection of vitamin D3 combined with high-fat feed.Blood lipid levels were measured for each group,including total cholesterol (TC),triglyceride (TG),low-density lipoprotein cholesterol (LDL-C),and high-density lipoprotein cholesterol,and the pathological change of the aorta was observed.The UPLC-Orbitrap MS technique was used to perform a lipidomic study of the plasma samples from each group.Results:Compared with the blank group,the model group had significant increases in the levels of TC,TG,and LDL-C (P<0.05 or P<0.01);compared with the model group,the administration group had significant reductions in the levels of TC,TG,and LDL-C (P<0.01).Histopathology showed that the administration group had a reduction in lipid deposition and an improvement in aortic intimal thickening.A total of 29 lipid subclasses were identified,with 959 lipid molecules in total.There were 62 differentially expressed lipid molecules between the administration group and the model group,mainly glyceryl phosphatide,sphingolipid,and glycerides.The changing trend of 22 differentially expressed metabolites in the administration group was contrary to that in the model group.Conclusion:Zhilong Huoxue Tongyu capsules can reduce blood lipids and improve atherosclerosis by acting on the main pathways of glyceryl phosphatide metabolism and glyceride metabolism.
