基于网络药理学探讨抗病毒颗粒治疗呼吸道感染的作用机制

摘要:
目的:通过网络药理学探讨抗病毒颗粒治疗呼吸道感染的作用靶点及信号通路,并阐述其作用机制。方法:运用TCMSP、TCMID及BATMAN-TCM数据库筛选抗病毒颗粒中中药的主要化学成分及作用靶点,通过UniProt和PubMed等数据库查询靶点对应的基因,使用DisGeNET数据库获取呼吸道感染相关靶点基因,运用Cytoscape 3.7.2软件构建药材-化合物-靶点(基因)-疾病网络,采用STRING数据库创建蛋白相互作用核心网络(PPI),再通过DAVID数据库进行GO和KEGG分析。结果:抗病毒颗粒中有槲皮素、木犀草素、汉黄芩素、山柰酚等89个活性成分,作用在PTGS2、TNF、AKT1、NOS2、IL-6等靶点蛋白上,通过肿瘤坏死因子(TNF)、核转录因子κB(NF-κB)、低氧诱导因子1(HIF-1)等信号通路发挥抗病毒的治疗作用。结论:抗病毒颗粒治疗呼吸道感染的作用机制可能与其活性化合物通过多靶点、多通路发挥抗病毒、抗炎、免疫调节等作用有关,这为进一步探讨其作用的物质基础以及作用机制提供了参考。

Abstract:
Objective:To investigate the action targets and signaling pathways for antiviral granules and its mechanism of action in the treatment of respiratory infection based on network pharmacology.Methods:TCMSP,TCMID,and BATMAN-TCM databases were used to screen out the main chemical components and action targets of the medicinal materials in antiviral granules,and UniProt and PubMed databases were used to obtain the corresponding genes of targets.Cytoscape 3.7.2 was used to construct a medicinal material-compound-target (gene)-disease network,STRING database was used to create a protein-protein interaction network,and DAVID database was used to perform gene ontology and Kyoto Encyclopedia of Genes and Genomes analyses.Results:There were 89 active components in antiviral granules,including quercetin,luteolin,wogonin,and kaempferol,which act on the target proteins such as PTGS2,TNF,AKT1,NOS2,and IL-6.Antiviral granules exerted an antiviral effect via multiple signaling pathways including tumor necrosis factor,nuclear factor-kappa B,and hypoxia-inducible factor-1.Conclusion:The mechanism of action of antiviral granules in the treatment of respiratory infection is possibly associated with the active compounds that exert antiviral,anti-inflammatory,and immunoregulatory effects via multiple targets and pathways,which provides a reference for further research on material basis and mechanism of action.
