基于网络药理学探讨芎芷地龙汤治疗偏头痛的作用机制

摘要:
目的:分析芎芷地龙汤的活性成分,预测其治疗偏头痛的作用靶点及信号通路。方法:通过TCMSP平台收集芎芷地龙汤活性成分并进行ADME筛选,收集活性成分潜在靶点;通过GeneCards数据库收集偏头痛的相关疾病靶点,并与药物活性成分靶点取交集得到共同靶点,进行PPI分析;使用Cytoscape构建“药物-靶点-疾病”网络并进行分析,通过DAVID数据库对共同靶点进行GO富集与KEGG富集分析。结果:共筛选出芎芷地龙汤85个潜在活性成分和145个潜在治疗靶点;芎芷地龙汤治疗偏头痛关键靶点为JUN、TNF、MAPK1、AKT1、RELA等;可能参与的通路有Pathway in cancer通路、HIF-1信号、TNF信号通路、TLR信号通路、神经活性配体-受体相互作用、钙信号通路、T细胞受体信号通路等。结论:槲皮素、β-谷甾醇等是芎芷地龙汤中的主要活性成分;TNF、MAPK1、AKT1等基因可能在治疗偏头痛时参与调控;芎芷地龙汤主要通过调节氧化应激反应、低氧反应、炎性因子释放、神经递质传递、血管张力等机制治疗偏头痛。

Abstract:
Objective:To investigate the active components of Xiongzhi Dilong decoction and its action targets and signaling pathways in the treatment of migraine.Methods:The TCMSP platform was used to collect the active components of Xiongzhi Dilong decoction and perform ADME screening,and the potential targets of these active components were collected.The GeneCards database was used to collect the disease targets associated with migraine,and the intersection of the disease targets and the targets of active components was performed to obtain common targets and perform a protein-protein interaction analysis.Cytoscape was used to construct a “drug-target-disease” network and perform related analyses,and the DAVID database was used to perform gene ontology enrichment analysis and Kyoto Encyclopedia of Genes and Genomes enrichment analysis.Results:A total of 85 potential active components and 145 potential treatment targets of Xiongzhi Dilong decoction were screened out.The key targets of Xiongzhi Dilong decoction in the treatment of migraine included JUN,TNF,MAPK1,AKT1,and RELA,and possible pathways involved included the Pathway in cancer pathway,HIF-1 signaling pathway,TNF signaling pathway,TLR signaling pathway,neuroactive ligand-receptor interaction,calcium signaling pathway,and T cell receptor signaling pathway.Conclusion:Major active components of Xiongzhi Dilong decoction include quercetin and β-sitosterol.The genes such as TNF,MAPK1,and AKT1 may participate in regulation during the treatment of migraine.Xiongzhi Dilong decoction exerts a therapeutic effect on migraine mainly by regulating oxidative stress response,hypoxia response,release of inflammatory factors,neurotransmitter transmission,and vascular tension.
