基于网络药理学探讨酸枣仁汤的作用机制

摘要:
目的:采用网络药理学方法探讨酸枣仁汤的作用机制。方法:借助TCMSP平台预测酸枣仁汤中药成分的作用靶点与通路,利用Cytoscape软件分别构建成分-靶点网络、PPI网络,使用DIVID数据库进行GO富集分析和KEGG信号通路富集分析。结果:经筛选获得酸枣仁汤中五味中药酸枣仁、川芎、知母、茯苓、甘草含有的135种活性成分,332个相应靶点;PPI网络包括293个节点;GO条目146个,其中生物过程为94个,分子功能条目34个,细胞组成条目18个;KEGG通路22条。结论:酸枣仁汤不仅可以用于治疗失眠、焦虑等精神系统疾病,还可以用于治疗肿瘤、循环系统等疾病,同时参与5-羟色胺、氮素代谢等生物过程,揭示了酸枣仁汤多成分、多靶点、多通路的药理作用特点。

Abstract:
Objective:To investigate the mechanism of action of Suanzaoren decoction based on network pharmacology.Methods:The TCMSP platform was used to predict the action targets and pathways of the components of Suanzaoren decoction.Cytoscape software was used to establish a component-target network and a protein-protein interaction (PPI) network.DIVID database was used to perform gene ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) signaling pathway enrichment analysis.Results:A total of 135 active components were obtained from Semen Ziziphi Spinosae,Rhizoma Chuanxiong,Rhizoma Anemarrhenae,Poria cocos,and Radix Glycyrrhizae in Suanzaoren decoction,with 332 corresponding targets in total.The PPI network had 293 nodes.There were 146 GO terms in total,with 94 terms of biological process,34 terms of molecular function,and 18 terms of cell component,and there were 22 KEGG pathways.Conclusion:Suanzaoren decoction can be used in the treatment of mental diseases including insomnia and anxiety,as well as tumor and circulatory system diseases,and it is also involved in the biological processes such as 5-hydroxytryptamine and nitrogen metabolization,suggesting that the pharmacological action of Suanzaoren decoction has the characteristics of multiple components,targets,and pathways.
