三金排石颗粒的质量标准研究

摘要:
目的:建立三金排石颗粒的质量标准。方法:采用薄层色谱法(TLC)分别对三金排石颗粒中的海金沙、栀子、大黄和山楂共四味中药进行定性研究;采用高效液相色谱法(HPLC)分别对金钱草中的槲皮素和山柰素进行定量研究,色谱柱为C18柱,流动相为甲醇-0.35%磷酸溶液(50∶50),流速为1.0 ml/min,检测波长为360 nm,柱温为30℃,进样量为10 μl。结果:所建立的TLC法中,海金沙、栀子、大黄和山楂斑点清晰,分离度好,专属性强。槲皮素和山柰素的检测质量浓度线性范围分别在0.010~0.100 mg/ml、0.010~0.101 mg/ml之间(r=0.9993、0.9998,n=6),精密度、稳定性、重复性试验的RSD均≤2%(n=6),平均加样回收率分别为101.86%、101.48%(n=6)。结论:本研究所建立的三金排石颗粒的质量标准,TLC法及HPLC法操作简便,准确快速,重现性好,可有效控制三金排石颗粒的质量。

Abstract:
Objective:To establish the quality standard for Sanjin Paishi granules.Methods:Thin-layer chromatography (TLC) was used to perform a qualitative study of Lygodium japonicum,Gardenia jasminoides,Rheum officinale,and hawthorn fruit in Sanjin Paishi granules;high-performance liquid chromatography (HPLC) was used to perform a quantitative study of quercetin and kaempferol in Lysimachia christinae on a C18 column with a mobile phase of methanol-0.35% phosphoric acid solution (50∶50) at a flow rate of 1.0 ml/min,a detection wavelength of 360 nm,a column temperature of 30℃,and a sample size of 10 μl.Results:In the TLC method established,Lygodium japonicum,Gardenia jasminoides,Rheum officinale,and hawthorn fruit had clear spots and were well separated with strong specificity.The mass concentrations of quercetin and kaempferol had linear ranges of 0.010~0.100 mg/ml and 0.010~0.101 mg/ml,respectively (r=0.9993 and 0.9998,n=6),and precision,stability,and reproducibility tests showed a relative standard deviation of ≤2% (n=6),with average recovery rates of 101.86% and 101.48%,respectively (n=6).Conclusion:The quality standard for Sanjin Paishi granules is established in this study,and both TLC and HPLC methods are simple,accurate,and rapid with good reproducibility and can effectively control the quality of Sanjin Paishi granules.
