基于网络药理学探析水陆二仙丹治疗糖尿病肾病蛋白尿的作用机制

摘要:
目的:基于网络药理学探讨水陆二仙丹治疗糖尿病肾病(DN)蛋白尿可能的作用机制及原理。方法:通过TCMSP平台检索获取水陆二仙丹中2味药的有效化学成分以及各化合物潜在作用靶点,利用GeneCards数据库筛选DN功能相关的靶标,将药物、有效成分、共同靶标导入Cytoscape 3.6.1。运用Cytoscape软件绘制“药物-疾病-靶点”可视化网络,利用软件分析功能得到主要药效成分及靶点。将药物作用靶点与DN功能相关靶点交集结果导入STRING10.5平台,构建“药物-靶标”蛋白相互作用(PPI)网络,利用DAVID数据库进行GO分析和KEGG富集分析。结果:挖掘出5种活性成分、116个相关靶点与本方治疗DN相互关联,其中PPI 网络图显示核心靶标AKT1、IL6、MAPK8、VEGFA、MAPK1等;KEGG 结果表明主要作用于AGE-RAGE信号等氧化应激相关通路。结论:网络药理学可科学预测水陆二仙丹治疗DN的主要作用机制,水陆二仙丹可能是通过对AGE-RAGE-NF-Кb/Ap-1调控轴为中心等多个靶点的抗氧化应激抑制作用控制DN蛋白尿。

Mechanism of action of Shuilu Erxian pills in treatment of diabetic nephropathy proteinuria based on network pharmacology

Abstract:
Objective:To investigate the mechanism of action and basic principle of Shuilu Erxian pills in the treatment of diabetic nephropathy (DN) proteinuria based on network pharmacology.Methods:The TCMSP platform was used to obtain the effective constituents of the two herbs in Shuilu Erxian pills and the potential targets of each compound,and the GeneCards database was used to screen out the targets associated with DN function.The drugs,effective constituents,and common targets were imported into Cytoscape 3.6.1.Cytoscape software was used to plot a “drug-disease-target” visualized network,and its analysis function was used to obtain the main effective constituents and targets.The intersection of drug targets and DN function-related targets were imported into STRING10.5 platform to construct a “drug-target” protein-protein interaction (PPI) network,and the DAVID database was used to perform GO analysis and KEGG enrichment analysis.Results:A total of 5 active components and 116 targets were found to be associated with this prescription in the treatment of DN.The PPI network showed that core targets included AKT1,IL6,MAPK8,VEGFA,and MAPK1,and the KEGG analysis showed that the active components mainly acted on oxidative stress pathways including AGE-RAGE signal.Conclusion:Network pharmacology can predict the main mechanism of action of Shuilu Erxian pills in the treatment of DN,and Shuilu Erxian pills may control DN proteinuria by exerting an anti-oxidative stress effect on multiple targets centered on the AGE-RAGE-NF-Кb/Ap-1 regulatory axis.
