目的:采用网络药理学研究分析牡丹皮-栀子药对治疗围绝经期综合征的多成分、多通路、多靶点的作用机制。方法:运用数据库检索得到牡丹皮-栀子药对的主要活性成分、相应作用靶点及围绝经期综合征的疾病靶点,用ConsensusPathDB数据库进行信号通路及基因本体论(GO)分析,并运用软件Cytoscape 3.7.0绘制关键成分-核心靶点网络图及蛋白互作网络图,软件imageGP绘制通路及GO分析气泡图。结果:得到包括堪非醇在内的关键活性成分17个,血管内皮生长因子(VEGF)A在内的核心靶点39个,GO分析得到包括细胞增殖调控在内的生物学过程970个;激素结合在内的分子功能116个;胰岛素受体复合体在内的细胞组分45个;富集到通路728条,涉及癌症通路和VEGF通路等。结论:牡丹皮-栀子药对可能通过调节激素水平、降低心血管疾病发生风险、改善胰岛素抵抗及调节癌症基因等靶点治疗围绝经期综合征。 |

Mechanism of action of Cortex Moutan Radicis-Fructus Gardeniae in treatment of perimenopausal syndrome based on network pharmacology |

Abstract: |
Objective:To investigate the multi-component,multi-pathway,and multi-target mechanism of action of Cortex Moutan Radicis-Fructus Gardeniae in the treatment of perimenopausal syndrome based on network pharmacology.Methods:Related databases were searched to obtain the main active components of Cortex Moutan Radicis-Fructus Gardeniae drug combination,corresponding targets,and the disease targets of perimenopausal syndrome.ConsensusPathDB database was used to perform signaling pathway and gene ontology (GO) analyses;the Cytoscape 3.7.0 software was used to plot the key component-core target network and the protein-protein interaction network;the imageGP software was used to plot the bubble charts of pathway and GO analyses.Results:A total of 17 key active components including kaempferol and 39 core targets including vascular endothelial growth factor-A were obtained,and GO analysis obtained 970 biological processes including regulation of cell proliferation.A total of 116 molecular functions were obtained including hormone binding,and 45 cellular components including insulin-receptor complex were obtained.The enrichment analysis obtained 728 pathways,involving cancer pathway and vascular endothelial growth factor pathway.Conclusion:Cortex Moutan Radicis-Fructus Gardeniae drug combination exerts a therapeutic effect on perimenopausal syndrome possibly by regulating hormone level,reducing the risk of cardiovascular disease,improving insulin resistance,and regulating cancer genes. |
