基于网络药理学探讨黄芪治疗糖尿病肾病的作用机制

摘要:
目的:探讨黄芪治疗糖尿病肾病的作用机制。方法:本研究通过中药系统药理学分析平台(TCMSP)数据库获取黄芪中含有的活性成分及成分所对应的靶点,通过GeneCards数据库挖掘糖尿病肾病疾病靶点,结合化合物及疾病靶点,构建成分-靶点网络图,并针对靶点进行基因本体(GO)富集分析及KEGG通路富集分析。结果:黄芪中含有 24个活性成分,对应糖尿病肾病110个靶点,可通过AGE-RAGE 信号通路、PI3K-AKT信号通路、丝裂原活化蛋白激酶(MAPK)信号通路、白细胞介素(IL)-17信号通路等调控糖尿病肾病增殖、凋亡、炎症等过程。结论:网络药理学研究表明黄芪可通过多种通路途经发挥作用,可为今后开展黄芪治疗糖尿病肾病相关实验验证提供依据。

Abstract:
Objective:To investigate the mechanism of action of Astragalus membranaceus in the treatment of diabetic nephropathy.Methods:The Traditional Chinese Medicine Systems Pharmacology database was used to obtain the active components of Astragalus membranaceus and their corresponding targets,and the GeneCards database was used to identify the targets of diabetic nephropathy.A component-target network was constructed based on the targets of the components and the disease,and a gene ontology enrichment analysis and a KEGG pathway enrichment analysis were performed for these targets.Results:There were 24 active components of Astragalus membranaceus,corresponding to 110 targets of diabetic nephropathy,and these components regulated the processes such as proliferation,apoptosis,and inflammation in diabetic nephropathy via the AGE-RAGE signaling pathway,the PI3K-AKT signaling pathway,the mitogen-activated protein kinase signaling pathway,and the interleukin-17 signaling pathway.Conclusion:The analysis based on network pharmacology shows that Astragalus membranaceus can paly a role via various pathways,which provides a basis for future experimental validation for Astragalus membranaceus in the treatment of diabetic nephropathy.
