星点设计-效应面法优选壮药盘龙止咳颗粒提取工艺的研究

摘要:
目的:通过星点设计-效应面法优选壮药盘龙止咳颗粒的最佳提取工艺,为后续研发更有效、更稳定、更方便的新制剂提供依据。方法:采用高效液相色谱法进行含量测定,采用单因素试验、星点设计-效应面法考察乙醇浓度、溶剂倍量、提取时间及提取次数,以山奈酚-3-O-龙胆二糖苷与岩白菜素总含量及干膏率的综合评分为评价指标,优选出壮药盘龙止咳颗粒最佳提取参数。结果:壮药盘龙止咳颗粒的最佳提取工艺为加入12倍量浓度为50%的乙醇,每次提取50 min,提取2次。结论:通过星点设计-效应面法优选盘龙止咳颗粒的提取工艺,方法预测性良好,重复性良好,简便可靠,为改变服用剂型研究提供了可靠依据。

Optimization of the extraction process for the Zhuang medicine Panlong Zhike granules using the central composite design-response surface method

Abstract:
Objective:To investigate the optimal extraction process for the Zhuang medicine Panlong Zhike granules using the central composite design-response surface method,and to provide a basis for further research and development of effective,stable,and convenient preparations.Methods:High-performance liquid chromatography was used for content determination,and the single-factor experiment and the central composite design-response surface method were used to evaluate alcohol concentration,times of solvent,extraction time,and times of extraction.The optimal extraction parameters for the Zhuang medicine Panlong Zhike granules were determined with the comprehensive score of the total content of kaempferol-3-O-gentiobioside and bergenin and dry extract yield as an assessment index.Results:The optimal extraction process for the Zhuang medicine Panlong Zhike granules was 12 times of 50% alcohol and extraction twice,with 50 minutes for each time.Conclusion:The central composite design-response surface method has good predictability and reproducibility and is convenient and reliable in optimizing the extraction process for the Zhuang medicine Panlong Zhike granules,which provides a reliable basis for the change in dosage form.
