基于网络药理学探讨化痰祛瘀汤治疗缺血性中风的潜在作用机制

摘要:
目的:基于网络药理学研究方法,探讨化痰祛瘀汤治疗缺血性中风的潜在作用机制。方法:运用TCMSP和相关文献记载检索筛选化痰祛瘀汤的主要活性化合物及相关作用靶点,通过Uniprot数据库获取作用靶点的标准名称,通过GeneCards数据库获取疾病的相关靶标,通过STRING数据库和Cytoscape 3.6.1软件构建与本病相关的核心靶点蛋白互作PPI网络。利用R语言软件对核心靶点进行GO功能富集分析和KEGG通路富集分析。结果:根据筛选条件得到化痰祛瘀汤207个有效化合物和176个药物相应靶点,根据平均度值7.43,筛选出核心靶点48个。GO功能富集分析得到条目89条(P<0.05),KEGG通路富集分析得到158条通路与本病有关(P<0.05)。化痰祛瘀汤中的活性化合物可能通过乙肝信号通路、MAPK信号通路、AGE-RAGE信号通路、TNF信号通路、IL-17信号通路、Th17细胞分化信号通路、C型凝集素受体信号通路、内分泌抵抗信号通路、细胞凋亡信号通路、Toll样受体信号通路等途径作用于JUN、MAPK3、MAPK1、TNF、IL6、AKT1、ESR1等核心靶点发挥其疗效。结论:化痰祛瘀汤与缺血性中风发病过程中的炎症反应、动脉粥样硬化、神经细胞凋亡、免疫反应等生物过程具有潜在的关联性与治疗作用。本研究为本药的临床应用及后续的效应机制研究提供了一定的理论基础。

Potential mechanism of action of Huatan Quyu decoction in treatment of ischemic stroke:An analysis based on network pharmacology

Abstract:
Objective:To investigate the potential mechanism of action of Huatan Quyu decoction in the treatment of ischemic stroke using the methods of network pharmacology.Methods:Traditional Chinese Medicine Systems Pharmacology and related articles were used to screen out the main active compounds of Huatan Quyu decoction and related targets.Uniprot database was used to obtain the standard names of targets,and GeneCards database was used to obtain related disease targets.STRING database and Cytoscape 3.6.1 software were used to establish the protein-protein interaction network of the core targets associated with this disease.R software was used to perform gene ontology (GO) enrichment analysis and KEGG pathway enrichment analysis of core targets.Results:A total of 207 effective compounds of Huatan Quyu decoction and 176 corresponding targets were obtained based on screening conditions,and 48 core targets were screened out based on a mean degree value of 7.43.GO enrichment analysis obtained 89 items (P<0.05),and KEGG pathway enrichment analysis obtained 158 pathways associated with this disease (P<0.05).Huatan Quyu decoction exerted a therapeutic effect by acting on the core targets including JUN,MAPK3,MAPK1,TNF,IL6,AKT1,and ESR1 via the hepatitis B signaling pathway,MAPK signaling pathway,AGE-RAGE signaling pathway,TNF signaling pathway,IL-17 signaling pathway,Th17 cell differentiation signaling pathway,C-type lectin receptor signaling pathway,endocrine resistance signaling pathway,cell apoptosis signaling pathway,and Toll-like receptor signaling pathway.Conclusion:Huatan Quyu decoction is associated with the biological processes such as inflammatory response,atherosclerosis,neural cell apoptosis,and immune response during the onset of ischemic stroke and thus exerts a therapeutic effect on this disease.This study provides a theoretical basis for clinical application and further research on mechanism of this drug.
