电针环跳、阳陵泉穴对神经痛大鼠海马区NAA、Glu变化的影响

摘要:
目的:从左侧海马区神经递质N-乙酰天门冬氨酸(NAA)、谷氨酸(Glu)变化探讨电针环跳、阳陵泉穴对神经痛的中枢作用机制。方法:将30只实验大鼠随机分为假手术组、模型组、电针组,每组各10只,建立慢性限制性损伤(CCI)神经痛大鼠模型。模型组造模后不干预;电针组造模后第7天选取右侧环跳、阳陵泉穴行电针干预1周;假手术组正常饮食不干预。采用Von-Frey测定法检测大鼠右侧足底机械痛阈值变化;磁共振波谱技术检测左侧海马区神经递质NAA、Glu的变化。结果:与假手术组相比,模型组造模后右侧足底机械痛阈值显著降低(P<0.01);左侧海马区Glu显著升高,NAA明显降低(P<0.01)。与模型组相比,电针组干预后右侧足底机械痛阈值显著提高(P<0.05),海马区Glu显著降低(P<0.05),而NAA明显升高(P<0.05)。结论:电针可缓解神经痛大鼠机械痛觉过敏,其作用机制可能与降低海马区Glu、提高NAA有关。

Effect of electroacupuncture at Huantiao and Yanglingquan acupoints on the changes in N-acetylaspartate and glutamic acid in the hippocampus of rats with neuralgia

Abstract:
Objective:To investigate the central mechanism of action of electroacupuncture at Huantiao and Yanglingquan acupoints by observing the changes in the neurotransmitters N-acetylaspartate (NAA) and glutamic acid (Glu) in the left hippocampus.Methods:A total of 30 experimental rats were randomly divided into sham-operation group,model group,and electroacupuncture group,with 10 rats in each group,and a rat model of neuralgia due to chronic constriction injury was established.The rats in the model group were given no intervention after modeling,and those in the electroacupuncture group were given electroacupuncture (beginning on day 7 after modeling) at Huantiao and Yanglingquan acupoints on the right side for one week.The rats in the sham-operation group were given normal diet without intervention.The von Frey test was used to measure the change in the mechanical pain threshold of the right hind paw,and magnetic resonance spectroscopy was used to measure the changes in the neurotransmitters NAA and Glu in the left hippocampus.Results:Compared with the sham-operation group,the model group had a significant reduction in the mechanical pain threshold of the right hind paw after modeling (P<0.01),as well as a significant increase in Glu and a significant reduction in NAA in the left hippocampus (P<0.01).Compared with the model group,the electroacupuncture group had a significant increase in the mechanical pain threshold of the right hind paw after intervention (P<0.05),as well as a significant reduction in Glu and a significant increase in NAA in the left hippocampus (both P<0.05).Conclusion:Electroacupuncture can alleviate mechanical hyperalgesia in rats with neuralgia,possibly by reducing Glu and increasing NAA in the hippocampus.
