血府逐瘀汤对大鼠深静脉血栓预防作用的实验研究

摘要:
目的:探讨血府逐瘀汤对大鼠深静脉血栓的预防作用。方法:将50只雄性SD大鼠随机分成血府逐瘀汤组、低分子肝素钠组、模型对照组和假手术组,每组予以不同用药,7d后除假手术组行假手术外,其余3组均建立深静脉血栓动物模型。造模术后72h各组均随机取5只大鼠脱颈处死,取股动脉血分离血浆,检测血浆组织型纤溶酶原激活剂(t-PA)和组织型纤溶酶原激活剂抑制物(PAI)水平;截取下腔静脉及分支静脉结扎段,电子天平称取血栓湿重。造模术后第10天将余下大鼠全部处死,同前方式检测t-PA和PAI水平及其凝血常规[包括血浆凝血酶时间(TT)、血浆凝血酶原时间(PT)、活化部分凝血酶时间(APTT)]、抗凝血酶Ⅲ(AT-Ⅲ)。结果:1)血栓湿重。低分子肝素钠组和血府逐瘀汤组之间差异无统计学意义(P>0.05),但2组与模型对照组相比血栓湿重均较轻,差异有统计学意义(P<0.05)。2)血浆t-PA及PAI。2个时段组内比较,血府逐瘀汤组和低分子肝素钠组的血浆t-PA、PAI水平差异无统计学意义(P>0.05),但2组与模型对照组相比,血浆t-PA含量均较高、PAI含量均较低,差异有统计学意义(P<0.01);2个时段之间对比,同组术后72h与术后10d t-PA及PAI水平无明显差异(P>0.05)。3)凝血指标。低分子肝素钠组和血府逐瘀汤组与模型对照组的TT、PT、AT-Ⅲ比较,差异均有统计学意义(P<0.05)。结论:血府逐瘀汤可减轻血栓湿重,在一定时间内促进t-PA及抑制PAI生成,从而可预防深静脉血栓。

Preventive effect of Xuefu Zhuyu decoction on deep venous thrombosis in rats:An experimental study

Abstract:
Objective:To investigate the preventive effect of Xuefu Zhuyu decoction on deep venous thrombosis in rats.Methods:A total of 50 male Sprague-Dawley rats were randomly divided into Xuefu Zhuyu decoction group,low-molecular-weight heparin sodium group,model control group,and sham-operation group,and these groups were treated with different drugs.After seven days of medication,the rats in the sham-operation group were given sham operation,and those in the other groups were used to establish a model of deep venous thrombosis.At 72 hours after modeling,5 rats each were selected from each group and were sacrificed by decapitation;plasma was isolated from femoral artery blood to measure the plasma levels of tissue plasminogen activator(t-PA)and tissue plasminogen activator inhibitor(PAI),and the legated segments of the inferior vena cava and its branches were collected to measure the wet weight of thrombus on an electronic balance.On day 10 after modeling,all the remaining rats were sacrificed,and the same method was used to measure the levels of t-PA and PAI,coagulation parameters [thrombin time(TT),prothrombin time(PT),and activated partial thromboplastin time(APTT)],and antithrombin III(AT-III).Results:There was no significant difference in the wet weight of thrombus between the low-molecular-weight heparin sodium group and the Xuefu Zhuyu decoction group(P>0.05),and these two groups had a significantly lower wet weight of thrombus than the model control group (P<0.05).The Xuefu Zhuyu decoction group and the low-molecular-weight heparin sodium group showed no significant changes in the plasma levels of t-PA and PAI at 72 hours and 10 days after modeling(P>0.05),but compared with the model control group,these two groups had a significantly higher plasma level of t-PA and a significantly lower plasma level of PAI(P<0.01);there were no significant changes in the plasma levels of t-PA and PAI in each group from 72 hours to 10 days after modeling(P>0.05).As for coagulation parameters,there were significant differences in TT,PT,and AT-III between the low-molecular-weight heparin sodium group/Xuefu Zhuyu decoction group and the model control group(P<0.05).Conclusion:Xuefu Zhuyu decoction can reduce the wet weight of thrombus,promote t-PA and inhibit PAI within a certain period of time,and thus prevent deep venous thrombosis.
